Significance Specialized cells in the inner ear translate sound into electrical signals for hearing, which are transferred to the brain through the cochlear nerve. Many of the molecular components of the inner ear are currently unknown. This paper uses a genetic approach to identify GRAP as a gene mutated in human deafness. In mice, Grap is present at the neuronal fibers innervating the auditory hair cells. The Drosophila homolog of the human Grb2-related adaptor protein (GRAP), drk, localizes to the synaptic terminals of neurons, and its disruption leads to hearing organ abnormalities associated with defects in locomotor behavior. We conclude that GRAP/drk plays an indispensable role in hearing.
